基于生信分析对国医大师刘尚义痹证药对豨莶草、海桐皮治疗骨关节炎靶基因的预测研究(临床研究)

【摘 要】目的:利用生物信息分析方法预测探索国医大师刘尚义痹证药对豨莶草、海桐皮治疗骨关节炎的靶点基因。方法:从公共基因芯片数据库OMIM筛选符合纳入条件的骨关节炎滑膜组织及软骨组织的基因组RNA芯片(样本均为骨关节炎患者和正常人的滑膜组织及软骨组织),利用GCBI在线实验室筛选出骨关节炎与正常组织的差异基因数据集Ⅰ;从BATMAN-TCM、TCMSP中药化合物数据库搜索豨莶草、海桐皮所含药物成分作用的效应基因数据集Ⅱ,分别将上述两个数据集及交集通过GCBI平台进行GO分析、KEGG分析以及参与生物功能的蛋白相互作用网络分析(MCODE分析)。结果:从符合研究纳入条件的3块骨关节炎患者与正常人滑膜组织基因芯片及1块软骨基因芯片数据集进行分析,最终得到102个差异性基因,从BATMAN中药化合物数据得到豨莶草及海桐皮药物组分调节基因共364个,取交集得出CASP3、PTGS2、JUN、PRKCA、PPARG与周围基因蛋白互作关系较为突出,之后的生物信息分析及相关文献证实这5个基因与炎症免疫反应及细胞凋亡联系密切。结论:豨莶草、海桐皮药对可能通过参与炎症及调节细胞凋亡相关通路来治疗骨关节炎。

Prediction of Target Genes of Classic Couplet Medicines Xixiancao and Haitongpi in the Treatment of Osteoarthritis by LIU Shang-yi:A Bioinformatics Analysis

【ABSTRACT】Objective:To predict and explore the target genes of classic couplet medicines Xixiancao(Herba Siegesbeckiae) and Haitongpi(Cortex Pittosporum) in the treatment of osteoarthritis by LIU Shang-yi using the bioinformatics analysis.Methods:Genomic RNA microarrays of synovial and cartilage tissues of osteoarthritis patients and normal subjects were screened from OMIM,a common gene chip database,and GCBI was used to screen the differential gene datasetⅠbetween osteoarthritis and normal tissues.The effect gene datasetⅡof Xixiancao and Haitongpi were searched from BATMAN-TCM and TCMSP Chinese Medicine Compound Databases.The two datasets were analyzed by GCBI platform for GO enrichment analysis,KEGG pathway analysis and MCODE analysis(a protein interaction network analysis).Results:A total of 102 deferentially expressed genes were obtained from three synovial tissue gene chips of osteoarthritis patients and normal subjects and a cartilage gene chip dataset.A total of 364 regulatory genes were obtained from the data of BATMAN Chinese herbal compounds.From the intersection,the relationship between CASP3,PTGS2,JUN,PRKCA,PPARG and peripheral gene proteins was more prominent.Bioinformatics analysis and related literature confirmed that these 5 genes were closely related to inflammatory immune response and apoptosis.Conclusion:Xixiancao and Haitongpi might treat osteoarthritis by participating in inflammation and regulating apoptosis-related pathways.
