独活寄生汤对膝骨关节炎大鼠软骨形态的影响（基础研究）

　　【摘　要】目的：观察独活寄生汤对膝骨关节炎大鼠软骨形态的影响，探讨其治疗膝骨关节炎的作用机制。方法：2月龄雄性SD大鼠80只，适应性喂养1周，随机分为空白对照组（20只）和造模组（60只）。造模组在第1，4，7天双膝关节腔注射0.2 mL质量分数为4%的木瓜蛋白酶。术后2周，拍摄膝关节小动物核磁共振成像（MRI）进行鉴定。造模成功后，随机分为模型对照组、独活寄生汤组和盐酸氨基葡萄糖胶囊组（阳性对照组），每组20只。HE染色和番红-固绿染色后，采用Mankin软骨组织学评分法进行评分。结果：MRI显示，空白对照组关节间隙正常，关节面平整、光滑；与空白对照组比较，模型对照组关节软骨变薄、局部有充盈性缺损，关节腔内积液明显增加。HE染色和番红-固绿染色结果表明，盐酸氨基葡萄糖胶囊与独活寄生汤都可以有效促进膝骨关节炎软骨修复，抑制软骨细胞凋亡，降低Mankin评分，但2组间差异无统计学意义（P > 0.05），组内8周时优于4周（P < 0.05）。结论：独活寄生汤能促进软骨生长与修复，保护膝骨关节炎模型大鼠膝关节的软骨形态，延缓软骨退变，从而起到治疗膝骨关节炎的作用。

Effects of Duhuo Jisheng Tang（独活寄生汤）on Cartilage Morphology in Experimental Rats with Knee Osteoarthritis

　　【ABSTRACT】Objective:To observe the effects of Duhuo Jisheng Tang（独活寄生汤）on cartilage morphology in experimental rats with knee osteoarthritis,to explore its mechanism in treating knee osteoarthritis.Methods:Eighty two-month-old SD rats,after adaptive feeding for one week,were randomly divided into a blank control group（20 rats） and a model group（60 rats）.On the 1,4,7 days,rats in the model group were injected with papain（0.2 mL,mass fraction being 4%）in their knee joint cavities.2 weeks after operation,magnetic resonance imaging（MRI）of the knee joint was taken for identification.After successful modeling,the rats were randomly divided into a model control group,a Duhuo Jisheng Tang group and a positive control group,20 in each group.After HE staining and safranin-fast green staining,the Mankin cartilage histological scoring method was used to score.Results:MRI showed that in blank control group,the articular space was normal and the articular surface was even and smooth.Compared with the blank control group,the articular cartilage of the model control group was thinner with local filling defect,and the intra-articular effusion increased significantly.HE staining and safranin-fast green staining showed that glucosamine hydrochloride capsule and Duhuo Jisheng Tang could effectively promote cartilage repair of knee osteoarthritis,inhibit chondrocyte apoptosis,and reduce Mankin score,but there was no significant difference between the two groups（P > 0.05）,and conditions within the group after 8 weeks was better than after 4 weeks（P < 0.05）.Conclusion:Duhuo Jisheng Tang can promote the growth and repair of cartilage,protect the cartilage morphology of knee joint in model rats with osteoarthritis,and delay cartilage degeneration,thus playing a role of treating knee osteoarthritis.
