肾阳虚对骨关节炎大鼠软骨退变的影响（基础研究）

　　【摘　要】目的：探索肾阳虚对骨关节炎（osteoarthritis,OA）大鼠软骨退变的影响。方法：将18只雄性SD大鼠随机分为正常组、OA组、OA肾阳虚组，每组6只。Hulth法建立OA模型，Hulth法加腺嘌呤灌胃建立OA肾阳虚模型。通过肾阳虚症状、体征评价表评价肾阳虚证候，光镜观察大鼠膝关节软骨组织病理学改变，OARSI和Mankin's评分评价软骨退变程度。结果：OA组和OA肾阳虚组大鼠均存在肾阳虚证候表现，且肾阳虚症状、体征积分均较正常组高（P = 0.002），其中OA肾阳虚组大鼠较OA组大鼠积分更高（P = 0.004）。OA组大鼠OARSI评分为4～6分、Mankin's评分属于OA早中期；OA肾阳虚组大鼠OARSI评分为12～16分、Mankin's评分属于OA中晚期，组间比较，差异有统计学意义（P < 0.01）。结论：OA大鼠存在肾阳虚证候表现，且肾阳虚明显加重OA大鼠的软骨退变程度。

Effects of Kidney Yang Deficiency on Cartilage Degeneration in Rats with Osteoarthritis

　　【ABSTRACT】Objective:To explore the effects of kidney yang deficiency on cartilage degeneration in osteoarthritis（OA）rats.Methods:Eighteen male SD rats were randomly divided into a normal group,an OA group and an OA kidney yang deficiency group,6 rats in each.The OA models were established by the Hulth method,and the kidney yang deficiency models were established by adenine gavage administration based on the Hulth method.The syndromes of kidney yang deficiency were evaluated by the symptom and sign evaluation table for kidney yang deficiency.The pathological changes of cartilage tissue were observed under the light microscope.The degree of cartilage degeneration was evaluated by OARSI and Mankin's scores.Results:Both of the OA group and the OA kidney yang deficiency group had the kidney yang deficiency syndrome,and the scores of kidney yang deficiency symptoms and signs was higher than those of the normal group（P = 0.002）,among which the score of the OA kidney yang deficiency group was higher than that of the OA group（P = 0.004）.The OARSI scores of the rats in the OA group were from 4 to 6.Mankin's score was in the early and middle stages of OA,and the OARSI scores of the rats in the OA kidney yang deficiency group was from 12 to 16,and the Mankin's score was in the middle and late stages of OA.The difference between groups was statistically significant（P < 0.01）.Conclusion:OA rats have the symptoms of kidney yang deficiency,and kidney yang deficiency can aggravate the degree of cartilage degeneration in OA rats.
