基于网络药理学探讨雷公藤-鸡血藤药对治疗类风湿关节炎的机制(临床研究)

  【摘 要目的:探讨雷公藤-鸡血藤药对治疗类风湿关节炎的作用机制。方法:使用中药系统药理学分析平台(TCMSP)筛选雷公藤-鸡血藤的活性成分,使用高通量基因表达数据库(GEO)、在线人类孟德尔遗传数据库(OMIM)、Genecards数据库筛选与类风湿关节炎相关的靶点,通过Cytoscape软件绘制化合物-靶点网络图。利用String数据库构建蛋白质相互作用(PPI)网络,利用R语言对靶点基因进行基因本体(GO)、京都基因与基因组百科全书(KEGG)生物通路富集分析。结果:筛选出木犀草素、山柰酚、雷公藤甲素、甘草查尔酮A70种活性成分,信号转导与转录活化因子3、丝裂原活化蛋白激酶1、丝裂原活化蛋白激酶14120个核心靶点,共同靶点主要涉及核受体的活动、乙型肝炎、前列腺癌、肿瘤坏死因子信号通路等1566个生物过程和152条信号通路。结论:对雷公藤-鸡血藤药对治疗类风湿关节炎的作用机制进行了初步探讨,为进一步阐述其作用机制奠定了基础。

Study on the Mechanism of the Drug Pair-Tripterygium Wilfordii and Caulis Spatholobi in the Treatment of Rheumatoid Arthritis Based on Network Pharmacology

  ABSTRACTObjective:To explore the mechanism of the drug pair-Tripterygium Wilfordii and Caulis Spatholobi in the treatment of rheumatoid arthritis.Methods:TCMSP was used to screen the active components of Tripterygium Wilfordii and spatholobus Spatholobi.Databases GEO,OMIM and Genecards were used to screen the targets related to rheumatoid arthritis.The "compound target" network diagram was drawn by the software Cytoscape.The PPI network was constructed using the database String.The target genes were enriched in GO and KEGG by R language.Results:Seventy active components such as Luteolin,Kaempferol,Triptolide and Licochalcone A,and one hundred and twenty core targets,including signal transducer and activator of transcription 3,mitogen activated protein kinase 1 and mitogen activated protein kinase 14 were screened.The common targets mainly involved 1566 biological processes,such as nuclear receptor activity,hepatitis B,prostate cancer,tumor necrosis factor signaling pathway and 152 signal pathways.Conclusion:The mechanism of Tripterygium Wilfordii and Caulis Spatholobi in the treatment of rheumatoid arthritis was primarily discussed,which laid a foundation for further elaboration for its mechanism.
