腺嘌呤制备肾虚骨关节炎模型最适浓度研究(基础研究)

  【摘 要目的:采用不同浓度腺嘌呤进行肾虚骨关节炎复合模型建模,建模成功后对其进行肾虚与骨关节炎各项指标测定,选择最适腺嘌呤建模浓度。方法:45只SD大鼠随机分为空白对照组,模型对照组,腺嘌呤高(150 mg·kg-1)、中(100 mg·kg-1)、低(50 mg·kg-1)剂量组,每组9只。造模成功后检测大鼠肾虚以及骨关节炎各项指标,包括大鼠证候评分、皮质酮(CORT)、雌二醇(E2)、睾酮(T)含量,关节大体情况,HE染色和Mankin's评分。记录大鼠死亡率。结果:腺嘌呤高剂量组大鼠证候评分高于腺嘌呤中剂量组与低剂量组(P < 0.05)。与空白对照组、模型对照组比较,腺嘌呤高、中剂量组E2E2/T、CORT均增高(P < 0.05),T降低(P < 0.01)。模型对照组和腺嘌呤高、中、低剂量组大鼠膝关节表面不光滑,潮线不完整,软骨厚薄不均匀。与空白对照组比较,模型对照组,腺嘌呤高、中、低剂量组Mankin's评分差异有统计学意义(P < 0.01)。腺嘌呤高剂量组死亡率为44%,其余4组无死亡。结论:腺嘌呤浓度100 mg·kg-1是最适肾虚骨关节炎复合模型建立浓度。

Study on the Optimal Concentration of Adenine in the Establishment of Osteoarthritis Model with Kidney Deficiency

  【ABSTRACTObjective:To establish compound models of osteoarthritis with kidney deficiency by using different concentrations of adenine.After the models are established successfully,the indexes of kidney deficiency and osteoarthritis were measured and the optimal concentration of adenine was selected.Methods:Forty-five SD rats were randomly divided into a blank control group,a model control group,groups with high adenine150 mg·kg-1,medium adenine100 mg·kg-1,low adenine50 mg·kg-1,9 rats in each.The indexes of kidney deficiency and osteoarthritis were measured after the models were established successfully,including syndrome score,cortisolCORT,estradiolE2,testosteroneT,general situation of joints,HE staining and Mankin's score.The mortality of rats was recorded.Results:The syndrome score of high dose group was higher than that of middle dose group and low dose groupP < 0.05.Compared with blank control group and model control group,E2,E2/T and CORT of high and medium dose adenine group were higherP < 0.05,while T was lowerP < 0.01.In the model control group and high,middle and low dose adenine group,the surface of knee joint was not smooth,the tide line was not complete,and the thickness of cartilage was not uniform.Compared with the blank control group,there was a significant difference in Mankin's score between the model control group and the high,medium and low dose adenine groupP < 0.01.The mortality rate of the high adenine group was 44%,and there were no death in the other four groups.Conclusion:100 mg·kg-1 is the best concentration for the establishment of the compound model of osteoarthritis with kidney deficiency.
