基于网络药理学和生物信息学研究麻黄附子细辛汤治疗类风湿关节炎的潜在机制(临床研究)

  【摘 要】目的:基于网络药理学和生物信息学挖掘麻黄附子细辛汤治疗类风湿关节炎的潜在作用机制。方法:采用中药系统药理学数据库和分析平台(TCMSP)筛选各味中药的入血活性成分和预测潜在的作用靶点,基于Gene Expression OmnibusGEO)数据库和DisGENET数据库检索类风湿关节炎相关的靶点,借助Cytoscape构建直接映射靶点的PPI网络,运用DIVID工具分析其GO分析和KEGG信号通路富集程度以研究其可能的分子机制。结果:通过TCMSP数据库检索到麻黄附子细辛汤相应化合成分362个,根据OB值和DL值筛选出入血活性成分35个,并预测潜在的作用靶点126个;采用R语言分析GEO数据库中编号为GSE55457的基因芯片筛选出明显的差异基因198个,检索DisGeNET数据库获得符合综合评分分值≥0.3的已知的疾病基因175个;借助GeneMANIA构建PPI网络表明关键靶点之间是相互作用;利用DAVID工具进行GO分析和通路富集分析显示,麻黄附子细辛汤干预类风湿关节炎的可能作用机制是复杂多样的,与白细胞介素(IL-6、肿瘤坏死因子、转录因子AP-1、血管内皮生长因子、γ干扰素及IL-1β6个靶点关系密切。结论:麻黄附子细辛汤干预类风湿关节炎具有多成分、多靶点的特点,是一种多通路、多层次、多因素相关联的最终结果。

On the Potential Mechanism of Mahuang Fuzi Xixin Tang(麻黄附子细辛汤)in the Treatment of Rheumatoid Arthritis Based on Network Pharmacology and Bioinformatics

  【ABSTRACTObjective:To explore the potential mechanism of Mahuang Fuzi Xixin Tang(麻黄附子细辛汤)in the treatment of rheumatoid arthritis based on network pharmacology and bioinformatics.Methods:TCMSP was used to screen the active components entering blood and predict the potential targets of each Chinese medicine.Based on the databases GEO and DisGENET,RA-related targets were retrieved.Cytoscape was used to build PPI network that directly maps targets,and DIVID was used to analyze its go analysis and KEGG signal path enrichment degree to study its possible molecular mechanism.Results:Three hundred and sixty-two components of Mahuang Fuzi Xixin Tang were retrieved with the database TCMSP,35 active components entering the blood were screened according to OB and DL values,and 126 potential targets were predicted.Through R language analysis,198 distinct genes were screened out from gene chip GSE55457 in GEO database and 175 known disease genes with comprehensive score over 0.3 were obtained from the database DisGeNET.PPI network constructed by GeneMANIA showed that the key targets interacted with each other.Go analysis and pathway enrichment analysis by DAVID showed that the possible mechanism of Mahuang Fuzi Xixin Tang for intervention of rheumatoid arthritis was complex and diverse,and closely related with the following six targets:IL-6,TNF,JUN,vascular endothelial growth factor,gamma interferon and IL-1β.Conclusion:The intervention of Mahuang Fuzi Xixin Tang has the characteristics of multi-component and multi-target,which is the final result of multi-channel,multi-level and multi factor correlation.
