基于肠道上皮细胞相关转运子探讨四妙散对高尿酸血症小鼠的祛湿机制(基础研究

  【摘 要】目的:探究四妙散对高尿酸血症小鼠回肠组织三磷酸腺苷结合盒转运蛋白转运体G2ABCG2)、葡萄糖转运蛋白9GLUT9)表达的影响。方法:36只小鼠随机分为空白对照组,模型对照组,四妙散低、中、高剂量组,别嘌醇组,每组6只。除空白对照组外,其余各组建立高尿酸血症小鼠模型,四妙散低、中、高剂量组分别给予低剂量(0.175 g·mL-1)、中剂量(0.35 g·mL-1)、高剂量0.7 g·mL-1)四妙散灌胃,别嘌醇组予别嘌醇(40 mg·kg-1)灌胃。检测各组小鼠血尿酸、回肠ABCG2GLUT9表达变化,探究四妙散治疗高尿酸血症的可能机制。结果:各模型组小鼠血清尿酸明显高于空白对照组(P < 0.05),不同剂量四妙散及别嘌醇治疗后小鼠血清尿酸水平降低,差异有统计学意义(P < 0.05),回肠ABCG2表达升高,GLUT9表达下降,差异有统计学意义(P < 0.05),其中四妙散中、高剂量组对ABCG2GLUT9影响最大。结论:四妙散通过调节回肠ABCG2GLUT9的表达起到降尿酸作用。

Study on the Dampness-eliminating Mechanism of Simiao San(四妙散)in Rats with Hyperuricemia Based on the Related Transporters of Intestinal Epithelial Cells

  【ABSTRACTObjective:To investigate the effect of Simiao San(四妙散)on the expressions of ABCG2 and GLUT9 in the ileum of mice with hyperuricemia.Methods:Thirty-six mice were randomly divided into a blank control group,a model control group,Simiao San low,medium and high dose groups,and an allopurinol group,6 mice in each group.In addition to the blank control group,hyperuricemia mice models were established in the other groups.The Simiaosan low,medium and high dose groups were correspondingly given intragastric administration of low dose0.175·g·mL-1,medium dose0.35 g·mL-1,and high dose0.7 g·mL-1of Simiao San,so did the allopurinol group with allopurinol40 mg·kg-1.The expressions of serum uric acid,ABCG2 and GLUT9 in ileum were detected to explore the possible mechanism of Simiao San in the treatment of hyperuricemia.Results:The level of serum uric acid in mice of each model group was significantly higher than that in the blank control groupP < 0.05.The serum uric acid level of mice treated with different doses of Simiao San and allopurinol decreased with statistical significanceP < 0.05.The expression of ABCG2 in ileum increased and the expression of GLUT9 decreased with statistical significanceP < 0.05.The Simiao San middle and high doses had the greatest impact on ABCG2 and GLUT9.Conclusion:Simiao San can reduce uric acid by regulating the expression of ABCG2 and GLUT9 in ileum.
