  【摘 要】目的:明确飞龙九一巴布贴的抗炎镇痛效果及其对膝骨关节炎滑膜炎的疗效。方法:采用角叉菜胶致大鼠足趾肿胀炎症模型,热板致小鼠舔足、冰醋酸致小鼠扭体2个疼痛模型及木瓜蛋白酶致新西兰大白兔膝骨关节炎滑膜炎模型等,观察飞龙九一巴布贴对大鼠足趾肿胀度、小鼠痛阈值、扭体次数、兔关节表面温度、关节周径、改良Lequesne MG得分及滑膜组织形态学等指标的影响。结果:①抗炎镇痛药效方面,与致炎组及致痛组比较,巴布贴组可显著降低大鼠足趾肿胀程度,提高小鼠痛阈值,减少小鼠扭体反应次数(P < 0.01)。在大鼠足肿胀实验中,致炎后0~2 h,巴布贴组对肿胀抑制作用弱于双氯芬酸二乙胺乳胶剂组(扶他林组)(P < 0.05);致炎后2~8 h,巴布贴组比扶他林组对肿胀的抑制作用更强(P < 0.05)。在小鼠热板法实验中,末次给药干预后0~1 h,巴布贴组对小鼠痛阈值的提高作用弱于扶他林组(P < 0.05);末次给药干预后1~3 h,巴布贴组比扶他林组对小鼠痛阈值的提高更显著(P < 0.05)。在小鼠扭体实验中,巴布贴组与扶他林组减少小鼠扭体次数的作用相当(P > 0.05)。②对膝骨关节炎滑膜炎影响方面,与空白组比较,模型组膝关节表面温度、关节周径、改良Lequesne MG得分显著升高(P < 0.01);与模型组比较,巴布贴组各指标均显著降低(P < 0.01),且滑膜炎症细胞浸润得到明显改善;与扶他林组比较,各指标差异均无统计学意义(P > 0.05)。结论:飞龙九一巴布贴抗炎镇痛作用显著,对膝骨关节炎滑膜炎亦有显著抑制作用。

Pharmacodynamic Study on the Anti-inflammatory and Analgetic Effect of Feilong Jiuyi Cataplasm(飞龙九一巴布贴) and Its Effect onKnee Osteoarthritis

  ABSTRACTObjective:To make clear the anti-inflammatory and analgesic effects of Feilong Jiuyi Cataplasm(飞龙九一巴布贴)and its effect on knee osteoarthritis and synovitis.Methods:Models of rat toe swelling and inflammation were induced by carrageenanthe inflammation-inducing group;two pain models respectively of licking feet and writhing body were induced by hot plate and by glacial acetic acidthe pain-inducing groups;and New Zealand white rabbits models with knee osteoarthritis synovitis were induced by papainthe papain group,to observe the effects of Feilong Jiuyi Cataplasm on the swelling degree of rat toe,pain threshold of mice,writhing times,temperature of rabbit joint surface,joint circumference and improved Lequesne MG score and synovial tissue morphology.Results:In terms of anti-inflammatory and analgesic effects,compared with the inflammation-inducing and pain-inducing groups,Cataplasm could significantly reduce the degree of toe swelling in rats,increase the pain threshold in mice and reduce the number of writhing reactions in miceP < 0.01.In the toe swelling experiment for rats,after two hours of inflammation,the inhibition on swelling in the Cataplasm group was weaker than that in the diclofenac diethylamine emulsion groupthe Votalin group)(P < 0.05;the inhibition on swelling in the Cataplasm group was stronger than that in the Votalin groupP < 0.05two to eight hours after inflammation.In the hot plate test for mice, after one hour the last administration intervention,the pain threshold of mice in the Cataplasm group was weaker than that in the Votalin groupP < 0.05;one to three hours after the last administration intervention,the pain threshold of mice in the Cataplasm group was higher than that in the Votalin groupP < 0.05.In the writhing test for mice,the effects on reducing the number of writhing in Cataplasm group and Votalin group were similarP > 0.05.Influences on knee osteoarthritis and synovitis:compared with the blank group,the knee surface temperature,joint circumference and improved Lequesne MG scores in the model group were significantly higherP < 0.01;compared with the model group,the indexes in the Cataplasm group were significantly lowerP < 0.01,and the synovial inflammatory cell infiltration was significantly improved;compared with the Votalin group,there was no significant difference in each indexP > 0.05.Conclusion:Feilong Jiuyi Cataplasm has significant anti-inflammatory and analgesic effects,and also has a significant inhibitory effect on knee osteoarthritis and synovitis.
