类风湿关节炎患者与药源性胃黏膜损伤的生物信息学分析(文献研究)

【摘 要】目的:利用生物信息学分析类风湿关节炎患者与药源性胃黏膜损伤的关系。方法:通过GeneCards、OMIM、TTD、pharmGKB、drugbank数据库检索“rheumatoid arthritis”“drug induce gastric mucosal injury”,获取类风湿关节炎及药源性胃黏膜损伤2个疾病基因,制作韦恩图获取2个疾病的共同基因,将共同基因导入STRING进行PPI网络分析、GO功能富集及KEGG通路分析。结果:通过检索不同数据库整理去重后,筛选出相关类风湿关节炎基因4953个,药源性胃黏膜损伤相关基因1489个,将2个疾病相关基因取交集后获得共同靶点基因1060个,PPI网络显示TNF、AKT1、GAPDH、CTNNB1、STAT3等为共同关键基因,GO富集主要与线粒体、腺苷受体信号通路、蛋白活性、磷酸酶结合等相关,KEGG信号通路包括NF-κB信号通路、JAK-STAT信号通路、Wnt信号通路等。结论:所获得的类风湿关节炎及药源性胃黏膜损伤的共同关键基因和涉及的信号通路,有助于理解两者在疾病过程中的相关性,为药物的研发提供理论参考。

Bioinformatics Analysis of Rheumatoid Arthritis Patients and Drug-induced Gastric Mucosa Injury

【ABSTRACT】Objective:To analyze the relationship between rheumatoid arthritis patients and drug-induced gastric mucosa injury by bioinformatics.Methods:Databases such as GeneCards,OMIM,TTD,pharmGKB,and drugbank were used to retrieve "rheumatoid arthritis" and "drug induced gastric mucosa injury" to obtain two disease genes of rheumatoid arthritis and drug-induced gastric mucosal injury.Wayne map was made to obtain common genes of the two diseases.Common genes were introduced into STRING for PPI network analysis,GO function enrichment,and KEGG pathway analysis.Results:After searching different databases,4953 genes related to rheumatoid arthritis and 1489 genes related to drug-induced gastric mucosa injury were screened out.1060 common target genes were obtained after intersection of two disease related genes.PPI network showed that TNF,AKT1,GAPDH,CTNNB1,STAT3 were common key genes.GO enrichment was mainly related to mitochondria,adenosine receptor signal pathway,protein activity,and phosphatase binding.The KEGG signaling pathway includes NF-κB signaling pathway,JAK-STAT signaling pathway,and Wnt signaling pathway.Conclusion:The acquired common key genes and involved signal pathways of rheumatoid arthritis and drug-induced gastric mucosa injury are helpful to understand their correlation in the disease process and provide theoretical reference for drug development.
