川乌、白芍治疗类风湿关节炎的网络药理学研究(临床研究)

【摘 要】目的:研究川乌、白芍治疗类风湿关节炎的作用机制。方法:以口服生物利用度(OB)≥30%、类药性(DL)≥0.18为指标,通过中药系统药理学数据库与分析平台(TCMSP)数据库分别获取川乌、白芍的活性成分、作用靶点,GeneCards数据库获取类风湿关节炎靶点,采用Cytoscape 3.7.2软件构建川乌、白芍活性成分−作用靶点网络,采用蛋白质相互作用数据库(String 10.0)构建蛋白-蛋白相互作用(PPI)网络,最后通过基因本体论(GO)及京都基因与基因组百科全书(KEGG)通路富集分析川乌、白芍防治类风湿关节炎的作用机制。结果:共得到川乌、白芍97个活性成分,涉及1041个作用靶点,通过筛选得出16种活性成分,检索到类风湿关节炎靶点3797个,其中川乌、白芍和类风湿关节炎的共有靶点34个,参与的KEGG通路有AGE-RAGE、白细胞介素(IL)-17、肿瘤坏死因子(TNF)、Toll样受体、p53、叉头框转录因子O(FOXO)、JAK-STAT、活化PI3K-Akt、核转录因子-κB等信号通路。结论:川乌、白芍可能通过IL-6、蛋白激酶、丝裂原活化蛋白激酶等靶点作用于AGE-RAGE、IL-17、TNF、Toll样受体、p53、FOXO等等多项细胞因子及多条信号通路发挥治疗类风湿关节炎的作用,提示川乌、白芍多成分、多靶点、多途径的特征,对未来研究川乌、白芍治疗类风湿关节炎具有重要意义。

Network Pharmacology of Chuanwu and Baishao in the Treatment of Rheumatoid Arthritis

【ABSTRACT】Objective:To study the mechanism of Chuanwu(Radix Aconiti)and Baishao(Radix Paeoniae Alba)in the treatment of rheumatoid arthritis.Methods:According to the indexes of OB≥ 30% and DL ≥ 0.18,the active components and targets of Chuanwu and Baishao were obtained by TCMSP database,and the rheumatoid arthritis targets were obtained by GeneCards database,software 3.7.2 was used to construct the protein-protein interaction(PPI)network,and finally,the mechanism of them in the prevention and treatment of rheumatoid arthritis was analyzed by Gene Ontology(GO)and Kyoto Encyclopedia of genes and genomes(KEGG)pathway enrichment.Results:A total of 97 active components of Chuanwu and Baishao were obtained,involving 1041 targets.Through screening,16 active components were obtained,and 3797 targets of rheumatoid arthritis were retrieved,including 34 targets of Chuanwu,Baishao and rheumatoid arthritis.AGE-RAGE,IL-17,TNF,Toll-like receptor,p53,FOXO,JAK-STAT,activation of P13K-Akt,nuclear factor-κB and other signaling pathways involved in KEGG.Conclusion:Chuanwu and Baishao may be related to IL-6 and protein kinase,mitogen activated protein kinase and other targets act on AGE-RAGE,IL-17,TNF,toll like receptor,p53,FOXO and other cytokines and multiple signaling pathways to play a role in the treatment of rheumatoid arthritis,suggesting that Chuanwu and Baishao have the characteristics of multi-component, multi-target and multi-channel, which is of great significance for the future study of Chuanwu and Baishao in the treatment of rheumatoid arthritis.
