干燥综合征基因差异谱的生物信息学分析(临床研究)

  【摘 要】目的应用生物信息学方法研究干燥综合征的差异基因,为干燥综合征的治疗提供新靶点。方法GEO数据库中下载并筛选干燥综合征相关的差异表达基因,利用STRING在线数据库构建蛋白与蛋白相互作用关系网络,运用Cytoscape软件筛选出关键基因,通过DAVID数据平台对差异基因进行基因本体论(GO)和京都基因与基因组百科全书(KEGG)富集分析。结果通过分析基因芯片GSE127952后获得406个差异基因,筛选出STAT1MX1IFIT1IFIT3ISG1510个关键基因;GO分析显示差异基因主要富集在正向调节细胞因子产生、淋巴细胞分化、T细胞活化等生物学过程;KEGG分析主要涉及细胞因子与细胞因子受体的相互作用、趋化因子信号通路、Toll样受体信号通路、T细胞受体信号通路等。结论上述所得的关键基因和信号通路可能与干燥综合征的疾病过程相关,为深入研究靶向治疗干燥综合征提供了理论参考。

Bioinformatics Analysis of Gene Differential Spectrum of Sjögren's Syndrome

  【ABSTRACTObjective:To study the differential genes of Sjögren's syndrome by bioinformatics,and to provide a new target for the treatment of Sjögren's syndrome.Methods:The deferentially expressed genes related to Sjögren's syndrome were downloaded and screened from the GEO database.The protein-protein interaction network was constructed by STRING online database.The key genes were screened out using the software of Cytoscape.The enrichment analyses of GO and KEGG for differential genes were carried out using DAVID data platform.Results:Four hundred and six deferentially expressed genes were obtained by analyzing gene chip GSE127952,and 10 key genes including STAT1,MX1,IFIT1,IFIT3 and ISG15 were screened out.Go analysis showed that the differential genes were mainly concentrated in biological processes such as positively regulating cytokine production,lymphocyte differentiation and T cell activation.KEGG analysis mainly involved the interaction between cytokines and cytokine receptors,chemotactic factor signaling pathway,Toll-like receptor signaling pathway,T cell receptor signaling pathway,etc.Conclusion:Above key genes and signal pathways may be related to the disease process of Sjögren's syndrome,which provides a theoretical reference for further research on the targeted treatment of it.
