乳糖凝集素-3在骨关节炎中作用的生物信息学分析

  【摘 要】目的:利用生物信息学方法分析半乳糖凝集素-3(gal-3)在骨关节炎中的作用机制。方法:GEO数据库中下载经gal-3处理的骨关节炎软骨细胞芯片,寻找芯片表达谱的差异基因并对其进行GO功能富集分析、KEGG通路富集分析以及疾病富集分析。此外,利用STRING数据库和Cytoscape软件构建差异基因蛋白互作网络,寻找网络结构中的核心基因。结果:共筛选出474个差异基因,其中包含246个上调的基因与228个下调的基因。GO富集分析结果表明,差异基因主要富集的生物学过程有骨髓白细胞迁移、白细胞趋化等;主要富集的细胞组分有细胞外基质、蛋白质细胞外基质等;主要富集的分子功能有细胞因子活性、趋化因子活性等。KEGG通路富集分析结果发现,差异基因主要涉及的通路有肿瘤坏死因子信号通路、白细胞介素-17信号通路等。疾病富集分析结果提示,差异基因主要富集的疾病有关节病、肺疾病、莱姆关节炎等。差异基因蛋白互作网络在Cytoscape软件共找出10个核心基因。结论:骨关节炎软骨细胞在gal-3作用下,可能通过上调白细胞介素-6等相关基因的表达在骨关节炎的进展中发挥重要作用。

Bioinformatics Analysis of the Role of Galectin-3 in Osteoarthritis

  【ABSTRACTObjective:To analyze the mechanism of Galectin-3Gal-3in osteoarthritis by bioinformatics.Methods:Gal-3-treated osteoarthritis chondrocyte microarrays were downloaded from the GEO database,finding out the differential genes of microarray expression profiles to carry out GO function enrichment analysis,KEGG pathway enrichment analysis and disease enrichment analysis.In addition,the interaction network of differential gene proteins was constructed using database STRING and software Cytoscape to find the core genes in the network structure.Results:A total of 474 differential genes were screened,including 246 up-regulated genes and 228 down regulated genes.The results of GO enrichment analysis showed that the main biological processes of differential gene enrichment were bone marrow leukocyte migration and leukocyte chemotaxis;the main cell components were extracellular matrix and protein extracellular matrix;the main molecular functions were cytokine activity and chemokine activity.The results of enrichment analysis for KEGG pathway showed that the main pathways involved in differential genes were tumor necrosis factor signaling pathway and interleukin-17 signaling pathway.The results of disease enrichment analysis indicated that the main diseases of differential gene enrichment were arthropathy,lung disease,Lyme arthritis and so on.Ten core genes were identified in the interaction network of differential gene  proteins by the software Cytoscape.Conclusion:Under the effect of Gal-3,osteoarthritis chondrocytes may play an important role in the development of osteoarthritis by up-regulating the expre-ssions of interleukin-6 and otherrelated genes.
