透骨消痛胶囊抑制大鼠骨关节炎软骨退变的实验研究

【摘 要】目的:探讨透骨消痛胶囊抑制骨关节炎软骨退变的机制。方法:将30只2月龄SPF级SD雄性大鼠按照随机数字表法分为空白对照组(10只)和模型组(20只)。模型组采用改良Hulth法建立大鼠骨关节炎模型,按照随机数字表法分为模型对照组和透骨消痛胶囊组,每组10只。透骨消痛胶囊组用透骨消痛胶囊溶液(10 mL·kg-1·d-1)灌胃,空白对照组、模型对照组连续等剂量生理盐水灌胃,持续12周。观察大鼠股骨髁软骨的整体形态学变化情况,另使用HE染色和甲苯胺蓝染色观察大鼠软骨形态结构及软骨蛋白多糖的表达,并用ELISA法检测滑膜组织中炎症相关因子肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β),以及基质金属蛋白酶-13(MMP-13)水平。结果:HE染色结果表明,空白对照组关节软骨表面光滑平整,软骨细胞排列规则,层次清晰;模型对照组软骨表面粗糙,软骨细胞排列紊乱,软骨细胞异常分化;透骨消痛胶囊组关节软骨表面比模型对照组完整,层次较为清晰,软骨细胞形态较好。甲苯胺蓝染色显示,空白对照组软骨染色呈紫红色,染色较深且均匀;模型对照组软骨染色明显变浅;透骨消痛胶囊组软骨染色较模型对照组深,染色相对均匀。ELISA法检测结果显示,模型对照组滑膜组织中TNF-α、IL-1β、MMP-13水平均高于空白对照组(P < 0.05),透骨消痛胶囊组低于模型对照组(P < 0.05),透骨消痛胶囊组与空白对照组比较,差异无统计学意义(P > 0.05)。结论:透骨消痛胶囊能有效抑制炎症反应所引起的关节软骨退行性病变,延缓骨关节炎病理进程。

Experimental Study on Tougu Xiaotong Jiaonang(透骨消痛胶囊)Inhibiting Cartilage Degeneration of Osteoarthritis in Rats

【ABSTRACT】Objective:To explore the mechanism of Tougu Xiaotong Jiaonang(透骨消痛胶囊)inhibiting cartilage degeneration in osteoarthritis.Methods:Thirty two-month-old SPF SD male rats were randomly divided into a blank control group(10 rats)and a model group(20 rats).The model group was divided into a model control group and a Tougu Xiaotong Jiaonang group randomly after the models were established according to modified Hulth method,with 10 rats in each group.For 12 weeks,the rats in the Tougu Xiaotong Jiaonang group were given Tougu Xiaotong Jiaonang solution(10 mL·kg-1·d-1)by gavage,while the blank control group and the model control group were given orally the same dose of saline by gavage.The morphological changes of their femoral condylar cartilage were observed.The morphological structure of rat cartilage and the expression of cartilage proteoglycan were observed by HE staining and toluidine blue staining.Inflammation-related factors such as TNF-α,IL-1β and MMP-13 in synovial tissue were detected by ELISA.Results:HE staining showed that the surface of articular cartilage in the blank control group was smooth,the arrangement of chondrocyte was regular and the layers were clear.The surface of cartilage in the model control group was rough,the arrangement of chondrocyte was disordered,and the chondrocyte was abnormally differentiated.The surface of articular cartilage in the Tougu Xiaotong Jiaonang group was complete,the layers were clear and the shape of chondrocyte was good,all better than those in the control group.Toluidine blue staining showed that the cartilage staining in the blank control group was purple red,deep and even.The cartilage staining in the model control group was significantly lighter.The cartilage staining in the Tougu Xiaotong Jiaonang group was deeper and more uniform than that in the model control group.ELISA results showed that the level of TNF-α,IL-1β and MMP-13 in synovial tissue of the model control group were higher than those of the blank control group(P < 0.05),and the level of them in the Tougu Xiaotong Jiaonang group were lower than those of the model control group(P < 0.05).There was no significant difference between the Tougu Xiaotong Jiaonang group and the blank control group(P > 0.05).Conclusion:Tougu Xiaotong Jiaonang can effectively inhibit the degenerative lesions of articular cartilage caused by inflammatory reaction and delay the pathological process of osteoarthritis.
