关于异型南五味子总三萜对类风湿关节炎大鼠
抗炎靶点干预的研究(基础研究)

【摘 要】目的:探讨异型南五味子总三萜对类风湿关节炎的抗炎效果、抗炎靶点和药物的作用机理。方法:将造模成功的胶原诱导性关节炎(CIA)大鼠100只随机分为模型对照组,甲氨蝶呤组,南五味子高、中、低剂量组,每组20只。通过观察CIA大鼠不同时间足趾肿胀容积程度,血清免疫球蛋白M(IgM)、IgM2RF、γ-干扰素(IFN-γ)水平改变,大鼠关节滑膜基质金属蛋白酶-1(MMP-1)、MMP-3表达情况。结果:在改善CIA大鼠足趾肿胀容积方面,甲氨蝶呤组、南五味子各剂量组12,15,30 d与模型对照组比较,差异有统计学意义(P < 0.01);给药15,30 d,南五味子高剂量组与甲氨蝶呤组比较,差异无统计学意义(P > 0.05)。大鼠血清中IgM、IgM2RF、IFN-γ含量比较,南五味子高、中剂量组较模型对照组均有显著抑制作用(P < 0.01),且高剂量组较甲氨蝶呤组对IgM、IgM2RF抑制差异有统计学意义(P < 0.01),低剂量组这3个指标有降低的趋势,但差异无统计学意义(P > 0.05)。在大鼠关节滑膜组织MMPs表达中,异型南五味子高、中、低剂量组均能显著抑制该蛋白高表达,与模型对照组和甲氨蝶呤组比较,差异有统计学意义(P < 0.05)。结论:异型南五味子各剂量对类风湿关节炎急性炎症期大鼠组织肿胀疼痛均有显著的缓解作用,且对炎性模型血清炎性细胞免疫因子和组织滑膜致炎蛋白酶有抑制效果,据此推测该药可能是通过阻断MMPs高表达抑制炎性因子及病情进展。

Study on the Interference of Total Triterpenes of Heterogenous Schisandra on Anti-Inflammatory Target in Rats with Rheumatoid Arthritis

【ABSTRACT】Objective:Discuss the anti-inflammatory effect and target of total three terpenes of fructus schisandrae chinensis on rheumatoid arthritis,and to infer the possible mechanism of the drug.Methods:One hundred CIA rats were randomLy divided into 5 groups,including model control group,methotrexate group,and South Schisandra group(high,medium,and low dose),20 rats in each group.The degree of swelling volume of the toes at different times after inflammation was observed.Serum IgM,IgMRF,IFN-γ level changes,CIA rat synovial MMP-1,MMP-3 expression of anti-triterpenoids and total anti-triterpenoids.Results:In improving the swelling volume of the CIA rat toes,the difference was statistically significant between 12,15,30 d in each dose group and model control group(P < 0.01).There was no significant difference between 15,30 d in the high dose group of South Schisandra and the methotrexate group(P > 0.05).For the comparison of IgM,IgMRF,and IFN-γ in rats serum,the high and medium dose of Schisandra were significantly inhibited compared with the model control group(P < 0.01),and the high dose group compared with the methotrexate group was significantly(P < 0.01).while the low dose group had a decrease in these three indicators,but there was no statistical difference(P > 0.05).In the expression of MMPs in rat joint synovial tissue,the high expression of the protein was significantly inhibited in the three groups of South Schisandra,which was statistically significant compared with the model control group and the methotrexate group(P < 0.05).Conclusion:Different dose groups of special-shaped South Schisandra had significant effects on the swelling and pain in acute inflammatory stage of rheumatoid arthritis,It also has inhibitory effect on inflammatory cytoimmune factor and synovial protease in serum of inflammatory model.It is assumed that the drug maybe by blocking the high expression inhibition factor of MMPs to achieve the progress of inhibition.
