膝关节炎治疗仪对SNP诱导体外大鼠软骨细胞凋亡的
影响(基础研究)

【摘 要】目的:观察膝关节炎治疗仪对硝普钠(SNP)诱导体外大鼠软骨细胞凋亡的影响。方法:采用膝关节炎治疗仪不同刺激强度干预1 mM SNP诱导第2代大鼠软骨细胞的凋亡模型,镜下观察软骨细胞形态,用噻唑蓝(MTT)法检测软骨细胞的活力,倒置荧光显微镜观察细胞核形态及密度变化。结果:倒置相差显微镜下观察膝关节炎治疗仪能减少细胞凋亡,MTT发现4级干预强度抑制细胞凋亡效果最佳,4,6-二脒基-2-苯基吲哚(DAPI)染色观察其能有效减轻软骨细胞核固缩。结论:膝关节炎治疗仪可能通过抑制软骨细胞凋亡从而达到防治骨关节炎的作用,但其具体抗凋亡途径有待进一步揭示。

Effect of Therapeutic Instrument for Knee Osteoarthritis on Apoptosis of Chondrocytes Induced by
SNP in Vitro

【ABSTRACT】Objective:To observe the effect of therapeutic instrument for knee osteoarthritis on apoptosis of chondrocytes induced by sodium nitroprusside(SNP)in vitro.Methods:The rat(GenerationⅡ)model with apoptosis of chondrocytes induced by 1 mM SNP in vitro was intervened with different stimulus intensities,observing the morphous of chondrocytes under microscope,detecting the vitality of chondrocyte with MTT,and observing the morphous of nucleus and the changes of density under inverted fluorescent microscopy.Results:Observed under the microscope,the therapeutic instrument for knee arthritis could reduce cell apoptosis.MTT showed that 4-scale intervention was the best intensity to inhibit apoptosis.According to DAPI(4',6-diamidino-2-phenylindole)staining,it could effectively reduce nuclear pyknosis.Conclusion:The therapeutic instrument for knee osteoarthritis can prevent and treat osteoarthritis by inhibiting apoptosis of chondrocytes,but the specific anti apoptotic pathway needs to be further revealed.
