加味独活寄生合剂对兔膝骨关节炎显微结构的影响(基础研究)

【摘 要】目的:观察加味独活寄生合剂对兔膝骨关节炎显微结构的影响。方法:采用随机数字表法将80只白兔分为正常对照组、模型对照组、阳性对照组和独活寄生组,每组20只。除正常对照组外,其余组均成功制备KOA模型。独活寄生组给予加味独活寄生合剂6.8 mL·kg-1·d-1灌胃,阳性对照组给予盐酸氨基葡萄糖胶囊0.07 g·kg-1·d-1灌胃,正常对照组和模型对照组给予等剂量生理盐水灌胃。在实验干预前、干预第2,4,8周时,对关节软骨形态及显微结构进行观察,并采用番红固绿染色进行Mankins关节软骨评分。结果:与模型对照组比较,独活寄生组膝关节大体观察得到有效改善。模型对照组、阳性对照组、独活寄生组Mankins软骨评分在干预前和干预第2,4,8周均高于正常对照组,且阳性对照组、独活寄生组Mankins软骨评分在干预第2,4,8周均低于模型对照组,差异有统计学意义(P < 0.05)。结论:加味独活寄生合剂可有效改善KOA关节软骨显微结构,这可能是其治疗KOA的作用机制。

Effect of Jiawei Duhuo Jisheng Heji(加味独活寄生合剂)on Microstructure of Rabbit Knee Osteoarthritis

【ABSTRACT】Objective:To observe the effect of Jiawei Duhuo Jisheng Heji(加味独活寄生合剂)on the microstructure of rabbit knee osteoarthritis(KOA).Methods:Eighty white rabbits were randomly divided into a normal control group,a model control group,a positive control group and a Duhuo Jisheng group,with 20 rabbits in each.KOA models were successfully prepared in all groups except the normal control group.The Duhuo Jisheng group was given 6.8 mL·kg-1·d-1 of Jiawei Duhuo Jisheng Heji by gavage,the positive control group was given 0.07 g·kg-1·d-1 of glucosamine hydrochloride capsule by gavage,and the normal control group and model control group were given the same dose of normal saline by gavage.The morphology and microstructure of articular cartilage were observed before the experimental intervention and at the 2nd,4th and 8th weeks of the intervention,and the Mankins articular cartilage score was performed by safranine solid green staining.Results:Compared with the model control group,by general observation the knee joint in the Duhuo Jisheng group was effectively improved,Mankins cartilage in model control group,positive control group,Angelica pubescens parasitic group.The scores were higher than those of normal control before intervention and at 2,4 and 8 weeks after intervention Group,and mankins cartilage score of positive control group and Angelica pubescens parasitic group.At 2,4,and 8 weeks of intervention,it was lower than that in the model control group,the difference being statistically significant(P < 0.05).Conclusion:Jiawei Duhuo Jisheng Heji can effectively improve the microstructure of KOA articular cartilage,which may be the mechanism of its treatment of KOA.
